Study of B cells populations in HIV- infected individuals

Abstract: Background: HIV infection is associated to abnormalities in lymphocyte populations, including B cells. These disturbances lead to an increase of specific co-infections and poor response to several vaccines. Therefore, characterization of B lymphocytes in HIV-infected patients is important to understand the viral induced-immune response. Objectives: The aim of this work was to characterize subpopulations of B lymphocytes in HIV-positive patients and correlate these findings with protein electrophoresis patterns found. Methods: 30 HIV-positive patients were included at different stages of progression under antriretroviral therapy. Subpopulations linfoctios B were immunotipyfied by flow cytometry as CD24 high CD38high (B transitional), CD27- (B virgin), CD27 + IgD + (B memory without isotype), CD27 + IgD (B memory isotype) and CD27 + (B total memory). Also, serum protein electrophoresis assays were performed using a semiautomatic analyzer and patterns were categorized as normal, hypogammaglobulinemia, polyclonal hypergammaglobulinemia, monoclonal or oligoclonal bands. Results: We found that compartment unbalances of B lymphocytes are mainly characterized by an increase in transitional B cell populations and decreased memory B cells without isotype switching. In addition, we found no correlation between the count of CD4 + T cells and B cell counts of total memory. Discussion: This study shows the compartment imbalance of B lymphocytes in adults infected with HIV even in patients under antiretroviral treatment. This has important implications for susceptibility to extracellular co-infections.
