Abstract: The ex vivo expansion of human hematopoietic stem and progenitor cells have great potential for clinical application in gene therapy, purging of contaminating malignant cells, graft engineering and the production of dendritic cells for tumor inmunotherapy. In the last two decades many reports have been published on this subject arguing that ex vivo cell expansion is feasible in different culture conditions ranging from statics cultures in gas permeable bags to continous perfusion cultures in bioreactors. In this review we will describe the basic methodology for cell culturing and expansion, and provide updated information regarding laboratory studies and clinical protocols applied to bone marrow, peripheral blood and umbilical cord blood cell populations.
